- Glycoproteins look like a small density on the membrane of viruses, such as the yellow-circled one below.
- In 3dmod, go to Edit → Object, and click “Type.
- A window like this will pop up.
- In “Points per contour”, make sure you input “2”, as we need 2 points to define one glycoprotein.
- Set the Mode to Model from Movie. When defining a glycoprotein, the first click (middle key on the mouse?) will be at the membrane (see the location of the green dot in the first picture). The second click (middle key on the mouse?) will be at the top tip of the glycoprotein (see the location of the red dot in the picture below.
- In this scenario, each “Contour” is each of your glycoproteins. Thus, you need to make sure you have only 2 “Points” per “Contour”.
- Save the model file (.mod) after you are done with one tomogram.
Note: The above example only applies to the side view picking. If you are using tomoStudio to pick top/bottom views, which only require one point per particle. You need to save the .mod separately by naming them *_side.mod or *_bottom.mod or *_top.mod
